为有效地研究烟草遗传多样性和筛选潜在的繁殖材料,以贵州省烟草科学研究院805份烟草种质为实验材料,分别在贵州省金沙和福泉2个试验点进行多点田间试验,并采用混合线性模型预测12个农艺性状的基因型效应值,以构建烟草的核心种质库并研究其遗传多样性.结果表明,待预测的12个农艺性状中,11个性状有显著的基因型效应,10个性状有显著的基因型与环境互作效应.利用预测的基因型值,比较不同抽样方法、抽样比率和系统聚类方法下核心子集多样性的结果表明,在10%抽样比率下,重心法聚类结合优先取样获得的S1C3_10子集成为烟草种质库的核心子集.通过主成分分析法比较核心种质与原始群体样本分布的空间结构和特征,运用相关系数检验原始群体性状间的遗传关系是否被核心种质很好地保留.结果表明,核心种质库能很好地代表烟草农艺性状的遗传多样性.一些潜在的高产育种材料如Y177和Y178也被包含在内.综上表明,作为种质资源的一个核心子集,烟草核心种质库的构建将有利于烟草育种者对整个资源的了解,促进遗传多样性在今后烟草育种计划中的利用.
In order to effectively study the genetic diversity of tobacco and screen potential breeding material, 805 tobacco germplasms of Guizhou Tobacco Research Institute were used as experimental materials to conduct multi-point field trials in two test sites of Jinsha and Fuquan in Guizhou Province respectively. Mixed linear models were used to predict the genotype effect values of 12 agronomic traits to construct a library of core collections of tobacco and to study their genetic diversity.The results showed that 11 of the 12 agronomic traits to be predicted had significant genotype effects , 10 traits had significant genotype-environment interaction effect.The results of comparing the diversity of core sub-sets under different sampling methods, sampling rate and system clustering method using the predicted genotype values showed that under the 10% sampling rate , And the core subset of tobacco germplasm library was obtained by clustering of the center of gravity method and the preferential sampling of S1C3_10.The core structure was compared with that of the original population by principal component analysis and the correlation coefficient was used to test the original population trait The results showed that the core genebank could represent the genetic diversity of tobacco agronomic traits well.One Potential high-yielding breeding materials, such as Y177 and Y178, are also included.In conclusion, as a core subset of germplasm resources, construction of tobacco core genebank will facilitate tobacco breeders’ understanding of the entire resource and promote genetic Use of diversity in future tobacco breeding programs.
