目的:建立运用波长切换技术同时测定黄连、枳实药对中柚皮苷、橙皮苷、巴马汀和小檗碱含量的液相色谱法。方法:采用Agilent C18(250mm×4.6mm,5μm)柱,以乙腈-0.3%磷酸0.2%三乙胺溶液为流动相进行梯度洗脱,检测波长分别切换为283nm,265nm。结果:柚皮苷、橙皮苷、巴马汀和小檗碱的线性范围分别为0.9392~4.696、0.4268~2.134、0.06544~0.3272和0.06832~0.3416μg,平均回收率分别为100.4%、99.4%、100.0%、101.2%,RSD分别为0.9%、2.4%、1.6%、2.9%。结论:本方法运用波长切换技术可同时测定黄连、枳实药对中柚皮苷、橙皮苷、巴马汀和小檗碱含量,具有快速、简便等特点。
OBJECTIVE: To establish a liquid chromatography method for the simultaneous determination of naringin, hesperidin, palmatine and berberine in Coptis chinensis and Fructus Aurantii by using wavelength-switching technique. Methods: Agilent C18 (250 mm × 4.6 mm, 5 μm) column was used with gradient elution of acetonitrile-0.3% phosphoric acid 0.2% triethylamine as the mobile phase. The detection wavelength was switched to 283 nm and 265 nm respectively. Results: The linear ranges of naringin, hesperidin, palmatine and berberine were 0.9392 ~ 4.696, 0.4268 ~ 2.134, 0.06544 ~ 0.3272 and 0.06832 ~ 0.3416μg respectively. The average recoveries were 100.4%, 99.4% 100.0%, 101.2%, RSD were 0.9%, 2.4%, 1.6%, 2.9% respectively. Conclusion: This method can be used to determine the contents of naringin, hesperidin, palmatine and berberine in Coptis chinensis and Fructus Aurantii Immaturus simultaneously with wavelength switching technology. The method is fast and simple.
