目的研究β-胡萝卜素(BC)和/或阿霉素(ADM)对裸小鼠人肝细胞癌组织模型(LTNM4)的治疗作用。方法第三次化疗后72小时处死全部裸鼠取血,采用酶联免疫吸附法(ELISA)检测血清甲胎蛋白(AFP),利用流式细胞仪(FCM)检测肿瘤及骨髓组织细胞周期分布情况。结果(1)BC单独使用对血清AFP水平无明显影响,当BC与ADM联合应用后,其血清AFP水平(2574.50ng/ml±431.53)较单独应用ADM(3727.03ng/ml±1498.67)显著降低(P<0.05)。(2)人肝癌细胞FCM检测结果BC使G0/G1%显著升高(P<0.01);ADM使G2M%显著升高(P<0.01);BC与ADM联合应用后,S%、G2M%显著升高(P<0.05,0.01)。(3)骨髓细胞FCM检测。BC对骨髓细胞周期分布无影响.ADM使S%、G2M%显著升高(均P<0.01).BC与ADM联合应用后,S%、G2M%较单用ADM显著降低(均P<0.05)。结论(1)ADM与BC联合应用对抑制LTNM4肝癌细胞生长及其AFP分泌具有协同作用,BC可显著增强化疗疗效;(2)BC可显著缓解ADM所致裸鼠骨髓损伤.对骨髓具有保护作用。
Objective To study the therapeutic effect of β-carotene (BC) and/or adriamycin (ADM) on human hepatocellular carcinoma tissue model (LTNM4) in nude mice. Methods Blood samples were taken from all nude mice at 72 hours after the third chemotherapy. Serum alpha-fetoprotein (AFP) was detected by enzyme-linked immunosorbent assay (ELISA). Cell cycle distribution of tumor and bone marrow was detected by flow cytometry (FCM). . Results (1) BC alone had no significant effect on serum AFP levels. When BC was combined with ADM, serum AFP levels (2574.50 ng/ml±431.53) were higher than ADM alone (3727.03 ng/ml±1498). .67) Significantly decreased (P<0.05). (2) FCM test results of human hepatocellular carcinoma cells increased G0/G1% significantly (P<0.01); ADM increased G2M% significantly (P<0.01); after BC and ADM were combined, S% G2M% increased significantly (P<0.05, 0.01). (3) FCM detection of bone marrow cells. BC has no effect on the cell cycle distribution of bone marrow. ADM increased the S% and G2M% significantly (both P<0.01). After the combination of BC and ADM, S% and G2M% were significantly lower than those of ADM alone (all P<0.05). Conclusion (1) The combination of ADM and BC has a synergistic effect on inhibiting the growth and AFP secretion of LTNM4 hepatocarcinoma cells. BC can significantly enhance the efficacy of chemotherapy. (2) BC can significantly alleviate bone marrow damage induced by ADM in nude mice. Has a protective effect on bone marrow.
