目的观察选择性环氧化酶-2抑制剂尼美舒利对人肝癌细胞(HepG2)生长及Smad4蛋白表达的影响。方法 MTS法检测不同浓度尼美舒利(25、50、100、200、400μmol/L)作用于HepG2后,对照组和实验组HepG2细胞增殖的变化;TUNEL法检测尼美舒利对HepG2凋亡的影响;荧光显微镜观察尼美舒利干预后,肝癌细胞Smad4蛋白表达的变化。结果尼美舒利对肝癌细胞HepG2的增殖有抑制作用,且随着尼美舒利浓度的增高,其对HepG2增殖的抑制作用逐渐增强;同时可以促进肝癌细胞核固缩、碎裂,诱导其凋亡;尼美舒利干预组肝癌细胞Smad4蛋白的荧光标记量明显高于对照组。结论尼美舒利可以通过上调肝癌细胞Smad4蛋白的表达来抑制肝癌细胞的增殖、促进其凋亡。
Objective To observe the effect of nimesulide, a selective cyclooxygenase-2 inhibitor, on the growth of HepG2 and the expression of Smad4 protein. Methods MTS assay was used to detect the proliferation of HepG2 cells in HepG2 cells treated with different concentrations of nimesulide (25,50,100,200,400μmol / L). The apoptosis of HepG2 cells was detected by TUNEL assay. The changes of Smad4 protein expression in hepatocellular carcinoma cells were observed by fluorescence microscope after nimesulide intervention. Results Nimesulide could inhibit the proliferation of HepG2 cells. With the increase of nimesulide concentration, the inhibitory effect on HepG2 proliferation was enhanced. At the same time, it could promote the nuclear condensation and fragmentation of HepG2 cells and induce their apoptosis In the nimesulide intervention group, the amount of fluorescent labeled Smad4 protein in hepatoma cells was significantly higher than that in the control group. Conclusion Nimesulide can inhibit the proliferation and promote the apoptosis of hepatoma cells by up-regulating the expression of Smad4 in hepatoma cells.
