目的:采用基因芯片技术研究理冲生髓饮对人卵巢浆液性乳头状囊腺癌细胞株SKOV3的作用机制。方法:Wistar大鼠随机分组,灌胃给药,采血制备含药血清。提取对照组及理冲生髓饮组肿瘤细胞mRNA,制备DNA探针并分别用Cy3和Cy5两种荧光染料标记,与基因表达谱芯片进行杂交,经扫描、分析,得出药物作用后表达有差异的基因。结果:共筛选出显著表达差异基因136种,其中16种基因表达水平下调,120种基因表达上调。结论:理冲生髓饮对卵巢癌细胞株SKOV3的生长有抑制作用,促进肿瘤细胞凋亡和抑制血管生成可能是其作用机制。
OBJECTIVE: To study the mechanism of “Li Chong Sheng Su Yin” on human ovarian serous papillary cystadenocarcinoma cell line SKOV3 by gene chip technology. Methods: Wistar rats were randomly divided into two groups: gavage and blood preparation. The mRNA of tumor cells was extracted from the control group and the Li Chong Sheng Su Yin Beverage Group. The DNA probes were prepared and labeled with Cy3 and Cy5 fluorescent dyes, respectively, and then hybridized with the gene expression microarray.
