目的:探讨Endostar联合放射治疗对Lewis肺癌小鼠移植瘤生长及介导氧跨膜转运的水通道蛋白1(AQP1)及乏氧诱导因子-1α(HIF-1α)mRNA表达的影响。方法:将32只Lewis肺癌移植瘤小鼠随机分为4组:空白对照组、单用Endostar组(ES组)、单用放疗组(RT组)、Endostar联合放疗组(ES+RT组),对各组肿瘤组织中AQP1及HIF-1αmRNA的表达情况进行检测。结果:AQP1mRNA的表达:与对照组相比,ES组变化无统计学差异(P>0.05)、RT组降低(P<0.05)、ES+RT组升高(P<0.05);HIF-1αmRNA的表达:与对照组相比,ES组变化无统计学差异(P>0.05)、RT组与ES+RT组均降低(P<0.05)。Pearson分析AQP1mRNA与HIF-1αmRNA的表达呈负相关(r=-0.58,P<0.01)。结论:Endostar联合放疗可能通过上调AQP1mRNA表达,增加细胞摄氧量,抑制HIF-1αmRNA表达,而增强射线对肿瘤细胞的杀伤作用。
Objective: To investigate the effects of Endostar combined with radiotherapy on the expression of aquaporin 1 (AQP1) and hypoxia inducible factor-1α (HIF-1α) mRNA in Lewis lung carcinoma xenografts and the transmembrane transport of oxygen. Methods: Twenty-two Lewis lung cancer xenografts were randomly divided into 4 groups: blank control group, endostar alone group (ES group), radiotherapy alone group (RT group), Endostar combined radiotherapy group (ES + RT group) The expression of AQP1 and HIF-1αmRNA in each tumor tissue was detected. Results: Compared with the control group, there was no significant difference in the expression of AQP1mRNA in ES group (P> 0.05), in RT group (P <0.05), in ES + RT group (P <0.05) Expression: Compared with the control group, there was no significant difference in ES group (P> 0.05), but both of RT group and ES + RT group decreased (P <0.05). Pearson analysis AQP1mRNA and HIF-1αmRNA expression was negatively correlated (r = -0.58, P <0.01). Conclusion: Endostar combined with radiotherapy may enhance the killing effect of radiotherapy on tumor cells by up-regulating the expression of AQP1 mRNA, increasing cellular oxygen uptake and inhibiting the expression of HIF-1α mRNA.
