目的 :研究OPG/RANK/RANKL在牙周炎联合血管钙化大鼠牙髓组织中的表达及其可能的作用。方法 :取36只SPF级雄性Wistar大鼠,随机分为4组,正常对照组(C组)、牙周炎组(CP组)、血管钙化组(VDN组)和牙周炎血管钙化联合组(CP+VDN组),并按照相应的方法建立动物模型。模型建立成功后,取含上颌第二磨牙的上颌骨,H-E染色观察牙髓组织的变化;免疫组织化学Envision法检测牙髓组织中OPG和RANKL蛋白质的表达及OPG/RANKL比值的变化。采用SPSS19.0软件包对数据进行统计学分析。结果:牙髓组织H-E染色观察显示,CP组、VDN组和CP+VDN组牙髓组织不同程度破坏,牙髓内可见中性粒细胞浸润,牙髓血管充血,成牙本质细胞空泡性变,牙髓坏死。其中,CP+VDN组最为显著,其次为CP组、VDN组。免疫组织化学染色结果显示,C组大鼠牙髓组织OPG强阳性表达,CP组、VDN组和CP+VDN组的平均吸光度值显著低于正常对照组(P<0.05),其中以CP+VDN组OPG阳性表达率最低;CP组、VDN组和CP+VDN组大鼠牙髓组织RANKL强阳性表达,各实验组的平均吸光度均显著高于C组(P<0.05),其中CP+VDN组RANKL阳性表达率最高;OPG/RANKL值C组最高,CP+VDN组最低,差异显著(P<0.05)。结论:牙周炎和血管钙化均对牙髓组织产生一定损伤,牙周炎合并血管钙化可加重这一损伤;OPG/RANKL/RANK可能在牙周炎和血管钙化对牙髓组织的影响中发挥重要作用。
Objective: To investigate the expression of OPG / RANK / RANKL in periodontitis combined with vascular calcification rat pulp tissue and its possible role. Methods: Thirty-six SPF male Wistar rats were randomly divided into 4 groups: normal control group (C group), periodontitis group (CP group), vascular calcification group (VDN group) and periodontitis vascular calcification group (CP + VDN group), and in accordance with the corresponding method to establish animal models. Conclusion: Both periodontitis and vascular calcification cause some damage to dental pulp tissue. Periodontitis and vascular calcification may aggravate this injury.
