观察Graves病(Graves disease,GD)、桥本甲状腺炎(hashimoto thyroiditis,HT)患者外周血调节性T细胞(regula-tory T cells,Treg)和叉头状转录因子FoxP3的表达情况及与抗甲状腺球蛋白抗体(TGAb)、抗甲状腺过氧化物酶抗体(TPOAb)表达的关系。用流式细胞术检测GD、HT患者外周血Treg、FoxP3的表达情况并与正常人群进行比较,并同时检测TGAb、TPOAb,观察FoxP3与TGAb、TPOAb之间的相关性。结果:GD组患者、HT患者CD4+CD25+Treg细胞数量与健康对照组相比无差异(P>0.05);而GD组患者、HT患者外周血中CD4+CD25+FoxP3+比例与对照组比较均存在着显著性差异(P<0.05)。对照组和AITD患者的TGAb、TPOAb水平存在显著性差异(P<0.05),且HT患者TGAb、TPOAb水平比GD组高(P<0.05)。Foxp3与TGAb(r=-0.623,P=0.000)、TPOAb(r=-0.555,P=0.000)成负相关。TGAb、TPOAb对于诊断和鉴别AITD具有重要意义,是HT诊断和鉴别诊断的首选指标。Treg及Foxp3在AITD的发生发展中起到重要作用,但Foxp3表达与AITD更有相关性。
To observe the expression of Treg and forkhead transcription factor FoxP3 in patients with Graves disease (GD) and Hashimoto thyroiditis (HT) and their relationship with anti-thyroid Globulin antibody (TGAb), anti-thyroid peroxidase antibody (TPOAb) expression. Flow cytometry was used to detect the expression of Treg and FoxP3 in peripheral blood of patients with GD and HT and compared with normal subjects. TGAb and TPOAb were also detected simultaneously to observe the correlation between FoxP3, TGAb and TPOAb. Results: The numbers of CD4 + CD25 + Treg cells in GD patients and HT patients were not significantly different from those in healthy controls (P> 0.05). However, the proportion of CD4 + CD25 + FoxP3 + in peripheral blood of GD patients and HT patients were There was a significant difference (P <0.05). The levels of TGAb and TPOAb in control group and AITD group were significantly different (P <0.05), and the levels of TGAb and TPOAb in HT group were higher than those in GD group (P <0.05). Foxp3 was negatively correlated with TGAb (r = -0.623, P = 0.000) and TPOAb (r = -0.555, P = 0.000). TGAb, TPOAb for the diagnosis and identification of AITD is of great significance, is the first choice of diagnosis and differential diagnosis of HT indicators. Treg and Foxp3 play an important role in the occurrence and development of AITD, but Foxp3 expression is more related to AITD.
