目的探讨调控微小RNAs(miRNAs)成熟通路相关基因与四川汉族人群胃癌易感的相关性。方法纳入207名四川汉族胃癌患者(病例组)和219名性别与年龄和病例组匹配的四川汉族人群(健康对照组)。运用高分辨率熔点曲线法(HRM)做基因分型,分别检测探讨调控mi RNAs成熟通路相关基因的2个SNP位点(DGCR8基因内的rs3757 G/A和AGO1基因内的rs636832A/G)的基因型和等位基因,比较2组人群基因及等位基因频率分布。结果病例组与对照组的基因分布均符合Hardy-Weinberg平衡;2个位点的基因型频率在病例组和对照组之间无明显差异:rs3757G/A为χ2=4.31,P=0.12;rs636832A/G为χ2=0.65,P=0.72。同时,这2个位点的等位基因频率在2组之间亦无明显差异:rs3757G/A为χ2=1.22,P=0.27,OR=1.22,95%CI为0.85-1.77;rs636832A/G为χ2=0.65,P=0.42,OR=0.88,95%CI=0.65为1.19。结论本组数据并未发现调控mi RNA成熟通路的相关基因的rs3757G/A和rs636832A/G位点的基因多态性与四川汉族人群胃癌的易感相关。
Objective To investigate the relationship between genes involved in the regulation of miRNAs mature pathways and susceptibility to gastric cancer in Sichuan Han population. Methods A total of 207 Chinese Han patients with gastric cancer (case group) and 219 matched Han and Han Chinese subjects (healthy control group) were enrolled in this study. High-resolution melting point curve (HRM) was used to genotype the two SNP loci (rs3757 G / A in DGCR8 gene and rs636832A / G in AGO1 gene) that regulate miRNAs mature pathway-related genes Genotype and allele, to compare the frequency distribution of genes and alleles in two groups of people. Results The genotype distributions of the two groups were in accordance with Hardy-Weinberg equilibrium. There was no significant difference in genotype frequencies between the two groups (rs3757G / A = χ2 = 4.31, P = 0.12; rs636832A / G is χ2 = 0.65, P = 0.72. At the same time, there was no significant difference in allele frequencies between the two groups: rs3757G / A was χ2 = 1.22, P = 0.27, OR = 1.22, 95% CI was 0.85-1.77; rs636832A / G was χ2 = 0.65, P = 0.42, OR = 0.88, 95% CI = 0.65 was 1.19. Conclusions Our data do not find the polymorphisms of rs3757G / A and rs636832A / G loci in the miRNA regulatory pathway and the susceptibility to gastric cancer in Sichuan Han population.
