目的测定流感病毒HA基因序列,从分子水平分析黑龙江省第1例甲型H1N1流感死亡病例病毒株以及2例2011年甲型H1N1流感病毒株HA基因的特点。方法采用MDCK细胞和Real-time PCR方法对甲型H1N1流感病毒HA基因片段进行分离、鉴定,并进行核苷酸序列测定和分析。结果将3例标本序列拼接后分别截取开放读码框内核苷酸同甲型H1N1代表株进行系统进化分析和同源性分析,结果显示新甲型H1N1流感病毒HA蛋白氨基酸与WHO推荐的疫苗株A/California/07/2009(H1N1)及中国的代表株A/Sichuan/1/2009(H1N1)亲缘关系最近,聚在一个分支上,同源性也较高,为98.6%~99.1%。结论目前的新甲型H1N1流感病毒的HA蛋白变异还很小,但应密切关注该病毒的变异情况。
Objective To determine the HA gene sequence of influenza virus and analyze the molecular characteristics of the first case of death from influenza A (H1N1) virus in Heilongjiang Province and the HA gene of two strains of H1N1 in 2011. Methods The HA gene fragment of influenza A (H1N1) virus was isolated and identified by MDCK cells and Real-time PCR. The nucleotide sequence was determined and analyzed. Results The nucleotide sequences of the three open reading frames were cloned and sequenced respectively. The phylogenetic analysis and homology analysis showed that the amino acid of HA protein of the new influenza A (H1N1) virus and the vaccine strains recommended by the WHO A / California / 07/2009 (H1N1) and China’s representative strain A / Sichuan / 1/2009 (H1N1) have recently been clustered in one branch and their homology is also high, ranging from 98.6% to 99.1%. Conclusion At present, the HA protein variation of the new influenza A (H1N1) virus is still small, but the variation of the virus should be closely monitored.
