目的:优化考察利用大孔吸附树脂纯化提取分离人工蛹虫草总核苷的工艺方法。方法:通过正交设计优化总核苷的提取分离条件,用高效液相色谱分析和检测各组分。结果:最佳提取条件为样品在10倍量水中80℃水浴提取3次,每次2 h,滤液合并,减压浓缩后上AB-8弱极性大孔树脂柱,蒸馏水洗脱除去多糖类大分子物质。以30%乙醇洗脱得总核苷提取液,减压浓缩,真空干燥后得到人工蛹虫草总核苷。结论:利用色谱法对提取过程中与提取液前体及中间体进行了比较分析,结果显示该工艺具有简单、快速、高效等优点。
OBJECTIVE: To optimize the process of purification and isolation of total nucleosides of Cordyceps militaris by macroporous adsorption resin. Methods: Optimize the conditions of extraction and isolation of total nucleosides by orthogonal design, and analyze and detect the components by high performance liquid chromatography. Results: The optimum conditions for the extraction of the sample in 10 times the amount of water 80 ℃ water bath extraction 3 times, each 2 h, the filtrate was combined, concentrated under reduced pressure AB-8 weakly polar macroporous resin column, distilled water to remove polysaccharides Macromolecular substances. The total nucleoside extract was eluted with 30% ethanol, concentrated under reduced pressure, and dried in vacuum to obtain artificial Cordyceps militaris total nucleosides. Conclusion: The chromatographic method was used to compare the extraction process with the extracts and intermediates. The results showed that the process was simple, rapid and efficient.
