目的 观察乳腺癌组织中NOEY2基因的mRNA表达情况 ,并探讨其与临床病理及其他分子指标的关系。方法 以RT PCR法和反义RNA探针原位杂交法 ,检测乳腺良恶性病变组织中NOEY2基因的mRNA表达。以免疫组化法检测乳腺癌石蜡标本中的雌激素受体 (ER)状态、Ki6 7标记指数以及p2 7和p2 1WAF1的表达。结果 RT PCR法检测显示 ,2 4例冻存乳腺组织中 ,6例良性病变NOEY2均为阳性 ;18例乳腺癌中 ,13例 (72 .2 % )NOEY2为阳性。原位杂交法检测显示 ,10例乳腺良性病变NOEY2均为阳性 ;而 6 0例乳腺侵袭性癌中 ,仅有 31例NOEY2为阳性 ,占 5 1.7%。良恶性病变NOEY2阳性率差异有显著性 (P <0 .0 2 5 )。NOEY2的阳性率有随组织学分级升高而递减的趋势。淋巴结阴性者阳性率为 76 .7% ,而阳性者仅为 2 6 .7% ,两者差异有显著性 (P <0 .0 0 1)。NOEY2基因mRNA表达与其他临床病理指标以及免疫组化指标均无显著相关性。结论 NOEY2基因可能在一定程度上参与了乳腺癌的发生和发展 ,该基因的失表达可能与乳腺癌的转移机制有关
Objective To observe the mRNA expression of NOEY2 gene in breast cancer and to explore its relationship with clinical pathology and other molecular markers. Methods RT-PCR and antisense RNA probe in situ hybridization were used to detect the mRNA expression of NOEY2 gene in benign and malignant breast lesions. The estrogen receptor (ER) status, Ki6 7 labeling index, and the expression of p2 7 and p2 1 WAF1 in breast cancer paraffin specimens were detected by immunohistochemistry. Results RT PCR showed that NOEY2 was positive in all 6 cases of benign breast lesions and in 13 cases (72.2%) of 18 breast cancers, NOEY2 was positive. In situ hybridization test showed 10 cases of benign breast lesions were positive NOEY2; and 60 cases of invasive breast cancer, only 31 cases of NOEY2 positive, accounting for 17.5%. There was a significant difference in the positive rate of NOEY2 between benign and malignant lesions (P <0.05). The positive rate of NOEY2 decreased with the increase of histological grade. The positive rate of lymph node negative was 76.7%, but the positive rate was only 26.7%, the difference was significant (P <0.01). NOEY2 gene mRNA expression and other clinicopathological parameters and immunohistochemical parameters were not significantly correlated. Conclusion NOEY2 gene may be involved in the occurrence and development of breast cancer to a certain extent. The loss of expression of this gene may be related to the metastasis of breast cancer
