对棉纤维作载体、对 - ( β-硫酸酯乙砜基 )苯胺 ( SESA)作活化剂固定化脂肪酶的工艺条件进行了研究 ,发现在醚化 p H值为 9.0~ 1 0 .0 ,交联 p H值为 7.0 ,酶的质量浓度为 6 mg· m L- 1,交联时间为 1 2 h,在室温条件下 ,得到最高固定化酶活力为 33U·g- 1。对上述条件下制备的固定化酶的稳定性进行了考察 ,用该固定化酶间歇水解橄榄油 ,重复使用 6次后相对水解率由 1 0 0 %降至 44%。该固定化脂肪酶最适使用的温度为 30~ 35℃ ,p H值为 9.0
The conditions for the immobilization of lipase on cotton fiber as carrier and the reaction of - (β-sulfoethylsulfone) aniline (SESA) as activator were studied. It was found that when the etherified p H value was 9.0 ~ 10.0, The crosslinked p H value was 7.0, the mass concentration of the enzyme was 6 mg · mL -1, the crosslinking time was 12 h, and the highest immobilized enzyme activity was 33 U · g -1 at room temperature. The stability of the immobilized enzyme prepared under the above conditions was investigated. With the immobilized enzyme, intermittent hydrolysis of olive oil was carried out, and the relative hydrolysis rate was reduced from 100% to 44% after being repeatedly used for 6 times.
