目的:对刺五加有效部位进行体外药效学评价。方法:采用MTT法检测细胞存活率,流式细胞仪测细胞凋亡率,紫外可见分光光度计测LDH的漏出量以及SOD、MDA、NO、NOS的活性和含量。结果:刺五加有效部位可以发挥良好的神经元保护作用,能提高MPP+损伤PC12细胞的存活率(P<0.01),降低细胞凋亡率(P<0.01),降低LDH的漏出量以及NO、NOS、MDA的含量和降低SOD活性,有统计学意义(P<0.05)。结论:刺五加有效部位对MPP+损伤PC12细胞具有一定的保护作用,其作用机制可能是通过降低PC12细胞NO、NOS、MDA的含量和LDH的漏出量,引起胞内SOD的活性增强,从而降低细胞凋亡率,提高细胞存活率而实现的。
Objective: To evaluate the pharmacodynamics of Acanthopanax senticosus in vitro. Methods: MTT assay was used to detect the cell viability, the rate of apoptosis by flow cytometry, the leakage of LDH and the activities and contents of SOD, MDA, NO and NOS by UV-vis spectrophotometer. Results: The effective part of Acanthopanax senticosus could play a protective role in neurons, improve the survival rate of PC12 cells (P <0.01), decrease the apoptosis rate (P <0.01), decrease the leakage of LDH, NOS, MDA content and reduce SOD activity, with statistical significance (P <0.05). CONCLUSION: The effective sites of Acanthopanax senticosus could protect PC12 cells against MPP + -induced injury. The mechanism may be that the activity of intracellular SOD is increased by decreasing the content of NO, NOS, MDA and the leakage of LDH in PC12 cells, Apoptosis rate, improve cell survival and achieve.
