目的 建立用高效液相色谱法测定血清苯妥英钠浓度的方法。方法 血清经乙醇抽提后 ,以甲醇为流动相 ,经色谱分离 ,在 2 4 0nm处检测 ,用标准曲线法进行定量分析。结果 苯妥英钠浓度在 0 .1~ 2 .0g/L范围内与峰面积之间具有较好的线性关系 ,回归方程为Y =0 .0 132 +1.0 736X ,相关系数 (r)为 0 .998,相对标准偏差 (RSD)为 0 .0 85% ,平均回收率为 99.53%。结论 此方法测定血清苯妥英钠浓度特异性强 ,简便可行 ,快捷灵敏。
Objective To establish a method for the determination of the concentration of phenytoin in serum by high performance liquid chromatography. Methods Serum was extracted by ethanol and methanol was used as mobile phase. After chromatographic separation, the serum was detected at 240 nm and quantified by standard curve method. Results There was a good linear relationship between the concentration of phenytoin sodium and the peak area in the range of 0.1-2.0 g / L, the regression equation was Y = 0.0131.0.0366X, the correlation coefficient (r) was 0.998 , The relative standard deviation (RSD) was 0.855% and the average recovery was 99.53%. Conclusion This method is specific and sensitive to the concentration of phenytoin in serum, which is simple and feasible.
