目的观察杠柳苷元(periplogenin)对人乳腺癌细胞MCF7的抑制作用并考察其抑制作用的机理。方法使用四氮唑盐(MTT)法测定杠柳苷元对MCF7细胞的增殖抑制效果,并使用蛋白免疫印迹法测定其中凋亡蛋白的表达。结果杠柳苷元对MCF7细胞起到了明显的抑制作用,其抑制作用呈剂量依赖性,蛋白免疫印迹法也观察到杠柳苷元促进MCF7细胞内凋亡相关蛋白的表达,其表达呈时间依赖性。结论杠柳苷元具有显著的体外抑制MCF7细胞增殖的作用,其抑制作用是通过促进MCF7细胞凋亡而发挥的。
Objective To observe the inhibitory effect of periplogenin on human breast cancer cell line MCF7 and investigate the mechanism of its inhibition. Methods The inhibitory effects of pheliotropin on the proliferation of MCF7 cells were determined by MTT method. The expression of apoptosis protein was detected by Western blotting. Results Perigunid -1 yuan significantly inhibited the proliferation of MCF7 cells in a dose-dependent manner. Western blotting also showed that perylene glycoside promoted the expression of apoptosis-related proteins in MCF7 cells in a time-dependent manner. Conclusion Peripheraldehyde (Glu) aglycone can significantly inhibit the proliferation of MCF7 cells in vitro, and its inhibitory effect is through promoting the apoptosis of MCF7 cells.
