目的采用RP-HPLC法测定马尿泡不同部位中东莨菪碱和阿托品的含量。方法采用Shimadzu VP C18柱(250mm×4.6mm,5μm),流动相为甲醇-水(30∶70,含0.02mol·L-1醋酸钠、0.1%三乙胺、0.3%四氢呋喃、冰醋酸调pH6.48),流速1.0mL·min-1,检测波长215nm,柱温30℃。结果东莨菪碱0.068~0.408μg与峰面积线性关系良好(r=0.9996);阿托品2.498~14.988μg与峰面积线性关系良好(r=0.9999)。结论所用方法简便、准确、灵敏度高、重复性好。
Objective To determine the contents of scopolamine and atropine in different parts of horse urine by RP-HPLC. Methods The mobile phase consisted of methanol-water (30:70) containing 0.02 mol·L-1 sodium acetate, 0.1% triethylamine, 0.3% tetrahydrofuran and glacial acetic acid using a Shimadzu VP C18 column (250 mm × 4.6 mm, .48), flow rate 1.0mL · min-1, detection wavelength 215nm, column temperature 30 ℃. Results Scopolamine showed a good linearity (r = 0.9996) between 0.068-0.408μg and peak area (r = 0.9996). Atropine 2.498-14.988μg had a good linear relationship with peak area (r = 0.9999). Conclusion The method is simple, accurate, sensitive and reproducible.
