目的:探讨低分子肝素对脂多糖诱导的人颊黏膜成纤维细胞分泌IL-8的影响。方法:原代培养正常人颊黏膜成纤维细胞,脂多糖刺激诱发炎症。不同浓度低分子肝素作用于炎症模型。采用ELISA方法检测细胞上清液中IL-8的变化。结果:脂多糖刺激后人颊黏膜成纤维细胞IL-8的表达高于正常对照组3 h(410.55 pg/mL与221.88pg/mL,P<0.05);低分子肝素组(1 U/mL)IL-8分泌量在6 h(552.85 pg/mL与885.81 pg/mL,P<0.05)、12 h(532.94 pg/mL与891.24 pg/mL,P<0.05)、24 h(405.96 pg/mL与887.88 pg/mL,P<0.05)较炎症组显著降低。结论:低分子肝素能显著抑制脂多糖刺激后人颊黏膜成纤维细胞IL-8的释放。
Objective: To investigate the effect of low molecular weight heparin on lipopolysaccharide-induced human buccal fibroblasts secrete IL-8. Methods: Primary cultured normal human buccal mucosal fibroblasts, lipopolysaccharide stimulation induced inflammation. Different concentrations of low molecular weight heparin on the inflammation model. ELISA method was used to detect the changes of IL-8 in the cell supernatant. Results: The expression of IL-8 in human buccal mucosa fibroblasts stimulated by lipopolysaccharide was significantly higher than that in the normal control group for 3 h (410.55 pg / mL and 221.88 pg / mL, P <0.05) The secretion of IL-8 was significantly higher at 6 h (552.85 pg / mL vs. 885.81 pg / mL, P <0.05), 12 h (532.94 pg / mL vs 891.24 pg / mL, P < 887.88 pg / mL, P <0.05) was significantly lower than the inflammation group. Conclusion: Low molecular weight heparin can significantly inhibit the release of IL-8 in human buccal fibroblasts stimulated by lipopolysaccharide.
