[目的]从脂质过氧化损伤阐述中药驱铅丸对慢性铅中毒大鼠肾保护作用的可能机制。[方法]在饮水中添加0.02%醋酸铅,连续60d以制作大鼠慢性铅中毒模型,然后将造模动物随机分为4组:中药高剂量治疗组(中药驱铅丸按3.5g.kg-1.d-1的剂量灌胃)、中药低剂量治疗组(驱铅丸按0.7g.kg-1.d-1的剂量灌胃)、EDTA治疗组(依地酸钠钙加普鲁卡因肌肉内注射,50mg.kg-1.d-1,连续注射4d为1个疗程,休息4d后进行下一个疗程,连续治疗7个疗程)及模型组(不予任何治疗),连续60d后观察治疗前后血、尿及肾脏的部分元素含量变化,并检测谷胱甘肽(GSH)、超氧化物歧化酶(SOD)、丙二醛(MDA)。[结果]经统计,与模型组比较,各治疗组尿铅、血铅含量显著降低(P﹤0.05),尿钙、尿锌降低,肾钙、肾锌增高(P﹤0.05),肾组织MDA降低,GSH增高(P﹤0.05,P﹤0.01),SOD变化没有显著性差异(P﹥0.05)。[结论]用于治疗慢性铅中毒的富锌中药驱铅丸能降低血液及肾组织铅含量,促进铅的排出;驱铅丸通过抗氧化损伤而发挥对铅造成的肾组织损害的保护作用。
[Objective] To explore the possible mechanism of the protective effect of traditional Chinese medicine and lead pill on renal protection of chronic lead poisoning rats from lipid peroxidation injury. [Methods] 0.02% lead acetate was added to drinking water for 60 days to prepare rat chronic lead poisoning model, and then the model animals were randomly divided into 4 groups: Traditional Chinese medicine high-dose treatment group (Chinese medicine to drive lead pill by 3.5g.kg-1 .d-1 dose gavage), Chinese medicine low-dose treatment group (Qigou pill administers 0.7 g.kg-1.d-1 gavage), EDTA treatment group (sodium etrigate calcium plus procaine) Intramuscular injection, 50mg.kg-1.d-1, continuous injection of 4d for 1 course of treatment, 4d after rest for the next course of treatment, 7 courses of continuous treatment) and model group (without any treatment), observed after 60d Changes in blood, urine, and kidney elements were measured before and after treatment, and glutathione (GSH), superoxide dismutase (SOD), and malondialdehyde (MDA) were measured. [Results] By statistics, compared with the model group, the urinary lead and blood lead levels in each treatment group decreased significantly (P<0.05), urinary calcium, urinary zinc decreased, renal calcium and renal zinc increased (P<0.05), kidney tissue MDA Decreased, GSH increased (P <0.05, P <0.01), there was no significant difference in SOD changes (P> 0.05). [Conclusion] Zinc-rich traditional Chinese medicine for treating chronic lead poisoning can reduce the lead content in blood and kidney tissue and promote the excretion of lead. The Quxie pill can protect the renal tissue damage caused by lead through antioxidant damage.
