了解各孕期胎盘组织中胰岛素样生长因子-Ⅰ(IGF-Ⅰ)的细胞定位,评估IGF-Ⅰ在胎儿、胎盘生长发育中的作用,采用免疫组化法对早、中、晚期妊娠胎盘组织中IGF-Ⅰ阳性细胞定位.结果显示:三个时期胎盘组织中IGF-Ⅰ主要位于绒毛小叶的合作细胞滋养层及细胞滋养层胞浆中。此外,中、晚期妊娠的Hoffbauer细胞及胎膜平滑绒毛层亦存在,但染色较以上两种细胞明显减弱,胎膜的羊膜细胞及蜕膜细胞偶尔可见阳性,胎盘蜕膜细胞则未见阳性着色.说明胎盘组织中IGF-Ⅰ主要由胎盘小叶的合体滋养层细胞、细胞滋养层细胞、Hoffbauer细胞及胎腹中的平滑绒毛膜细胞合成和分泌后作用于胎儿、胎盘组织,从而促进胎儿生长发育。
To understand the cellular localization of insulin-like growth factor-I (IGF-I) in placenta during pregnancy and to evaluate the role of IGF-I in the growth of fetus and placenta. Immunohistochemistry was used to detect the expression of IGF- IGF-I positive cells. The results showed that in the three stages of placenta, IGF-Ⅰ mainly located in the co-operative cytotrophoblast and cytotrophoblast cytoplasm of villous lobules. In addition, Hoffbauer cells and placental smooth villi of middle and late pregnancy also existed, but the staining of the two cells was obviously weaker than that of the above two groups. Occasionally, positive staining of amniotic membrane and decidual cells was observed in fetal membranes and positive staining of placental decidual cells was not seen . Explain placental IGF-Ⅰ mainly by the placental syncytiotrophoblasts, trophoblast cells, Hoffbauer cells and smooth fetal chorionic cells in the fetal synthesis and secretion of the role in the fetus, placenta, thereby promoting fetal growth and development.
