[目的]观察β-榄香烯诱导人骨肉瘤MG-63细胞的凋亡作用,并探讨其可能的机制.[方法]将不同质量浓度的β-榄香烯作用于人骨肉瘤MG-63细胞,用MTT法测定细胞存活率,流式细胞术测定细胞凋亡,免疫荧光染色、蛋白印迹检测凋亡相关蛋白的表达.[结果]与对照组比较,β-榄香烯处理组细胞存活率明显降低(P<0.05);凋亡相关蛋白表达明显增加(P<0.01).[结论]β-榄香烯可诱导人骨肉瘤MG-63细胞的凋亡,β-榄香烯诱导的细胞凋亡是通过线粒体途径的,同时激活caspase依赖性凋亡及caspase非依赖性凋亡2条凋亡途径.
[Objective] To observe the apoptosis effect of β-elemene on human osteosarcoma MG-63 cells and to explore its possible mechanism. [Methods] β-elemene with different mass concentrations was applied to human osteosarcoma MG- Cell viability was measured by MTT assay, apoptosis was detected by flow cytometry, immunofluorescence staining and Western blotting were used to detect the expression of apoptosis-related proteins. [Results] The cell viability in β-elemene treatment group was significantly higher than that in control group (P <0.01). [Conclusion] β-Elemene induces the apoptosis of human osteosarcoma MG-63 cells and the apoptosis induced by β-elemene Is a mitochondrial pathway that activates two apoptotic pathways, caspase-dependent apoptosis and caspase-independent apoptosis.
