目的:研究人胶质母细胞瘤(GBM)源性微血管内皮细胞(GBMDMEC)和正常人脑组织源性微血管内皮细胞(GBMDMEC)中Nogo-B的功能表达,了解Nogo-B在GBM血管生成中起的作用.方法:通过显微外科神经手术获得14例GBM标本,利用CD105免疫磁珠内皮细胞分选系统纯化内皮细胞,采用免疫组化染色,Real-time PCR和WesternBlot等方法检测其中Nogo-B在蛋白和mRNA水平上的表达.结果:与正常脑组织源性微血管内皮细胞相比较,Real-time PCR和Western Blot检测结果发现,在GBM源性微血管内皮细胞中Nogo-B表达上调.结论:Nogo-B在恶性GBM中mRNA和蛋白水平上表达均显著上调,证明Nogo-B参与了GBM新生血管形成,与GBM发生发展密切相关.
OBJECTIVE: To study the functional expression of Nogo-B in human glioblastoma (GBM) -derived microvascular endothelial cells (GBMDMEC) and normal human brain tissue-derived microvascular endothelial cells (GBMDMEC), and to understand the role of Nogo-B in GBM angiogenesis .Methods: 14 cases of GBM were obtained by microsurgical nerve operation, and endothelial cells were purified by CD105 immunomagnetic bead endothelial cell sorting system.Immunohistochemistry, Real-time PCR and Western Blot were used to detect the expression of Nogo- B expression at protein and mRNA levels.Results: Compared with normal brain tissue-derived microvascular endothelial cells, Real-time PCR and Western Blot results showed that Nogo-B expression was up-regulated in GBM-derived microvascular endothelial cells.Conclusions : Nogo-B in malignant GBM mRNA and protein levels were significantly increased, indicating that Nogo-B is involved in GBM neovascularization, closely related to the occurrence and development of GBM.
