血吸虫的生活史复杂,各发育阶段的生存环境和温度各不相同。热休克基因在适应环境压力和发育过程中活化,因而研究血吸虫的hsp70基因族有一定的意义。作者以λEMBL4载体构建血吸虫成虫DNA基因组文库。未扩增的文库用与hsp70编码区互补的23-mer寡核苷酸片段筛选出能与23-mer探针杂交含有一个3.5kbEcoRI片段的p.11克隆,该片段被SalⅠ,PvnⅡ,RsaⅠ,HaeⅢ酶分解,再将限制性片段亚克隆于M13测序载体中。从35个M13亚克隆的序列资料得出了一明确的结构序
The history of schistosomiasis is complex, living environment and temperature at different developmental stages vary. Heat shock gene activation in the adaptation to environmental stress and development, so the study of schistosoma hsp70 gene family has a certain significance. The authors constructed the DNA genome library of schistosomiasis with λEMBL4 vector. Untreated Libraries The pll clone containing a 3.5 kb EcoRI fragment hybridized to the 23-mer probe was screened with a 23-mer oligonucleotide fragment complementary to the hsp70 coding region and ligated with a SalI, PvnII, RsaI, HaeIII enzyme decomposition, and then the restriction fragment was subcloned in the M13 sequencing vector.
