目的探讨香椿子正丁醇提取物对脑缺血再灌注介导大鼠多器官功能障碍综合征的保护作用。方法大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型。动物随机分为假手术组、缺血模型组、溶媒对照组、香椿子正丁醇提取物(20mg/kg.d,30mg/kg·d)治疗组、阿司匹林阳性对照组。手术后断头取脑,行神经行为学评分判断神经功能损伤程度,称重法测量脑组织含水量。取肝和肺组织,分光光度法测定丙二醛(malondialdehyde,MDA)、一氧化氮(nitrogen monoxidum,NO)含量和超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧物酶(glutathione peroxidase,GSH-Px)和黄嘌呤氧化酶(xanthine oxidase,XO)活力的变化。结果 1香椿子正丁醇提取物可以改善脑缺血再灌注大鼠神经行为学评分(P﹤0.05)、降低脑缺血再灌注大鼠脑组织含水量(P﹤0.05),从而起到神经保护作用;2香椿子正丁醇提取物可减少肝和肺组织MDA和NO的生成(P﹤0.05)、抑制其SOD、GSH-Px、XO活力升高(P﹤0.05)。结论香椿子正丁醇提取物可通过其抗氧化应激效应对脑缺血介导的多器官功能障碍综合征发挥保护作用。
Objective To investigate the protective effect of n-Butyl alcohol extract of Toona sinensis on multiple organ dysfunction syndrome induced by cerebral ischemia-reperfusion in rats. Methods The focal cerebral ischemia-reperfusion model was established by middle cerebral artery occlusion in rats. Animals were randomly divided into sham operation group, ischemia model group, vehicle control group, Toona sinensis n-butanol extract (20mg / kg.d, 30mg / kg · d) treatment group, aspirin positive control group. After the operation, the brain was decapitated and the neurobehavioral score was used to judge the degree of neurological injury. The weight of brain tissue was measured by weighing method. The liver and lung tissues were taken out and the content of malondialdehyde (MDA), nitrogen monoxidum (NO) and superoxide dismutase (SOD), glutathione peroxidase (glutathione peroxidase, GSH-Px) and xanthine oxidase (XO) activity changes. Results 1 Toona sinensis n-butanol extract can improve the neurobehavioral scores of cerebral ischemia-reperfusion rats (P <0.05), decrease the water content of brain tissue in rats with cerebral ischemia-reperfusion (P <0.05) (P <0.05), and inhibited the activities of SOD, GSH-Px and XO in liver and lung tissue (P <0.05) .2. CONCLUSION N-Butyl alcohol extract of Toona sinensis plays a protective role against multiple organ dysfunction syndrome mediated by cerebral ischemia through its anti-oxidative stress effects.
