Optimization of Extraction Process of Compound Traditional Chinese Medicine by Orthogonal Design

[Objective] The objective was to screen the best extraction process of compound traditional Chinese medicine that is composed of Radix Sophorae Flavescentis,Rhizoma Smilacis glabrae and Cortex Dictamni and provide experimental evidence for the development and utilization of the compound prescription.[Method] The minimal inhibitory concentration(MIC) of extracts on Candida albicans as index,the orthogonal test was designed with 4 factors:concentration of ethanol(A),extraction time(B),the proportion of three components(C) and material-liquid ratio(D) to screen the optimum extraction conditions.[Result] The optimum technology was as follows:extraction solution of 70% ethanol(V/V),proportion of Radix Sophorae Flavescentis,Rhizoma Smilacis glabrae and Cortex Dictamni was 2:2:1,material-liquid ratio of 1:10 and extraction time of 2 h,under the above conditions the yield of the compound traditional Chinese medicine was 23% and the MIC of Candida albicans was 0.195 mg/ml.[Conclusion] The optimum extraction technology was obtained from the orthogonal test,which would provide reference for the development and utilization of the compound traditional Chinese medicine.
