目的 通过研究重组人白介素11(rhIL -1)对照射小鼠小肠上皮细胞凋亡及相关基因表达的影响,探讨IL-11在急性放 射病小鼠小肠上皮细胞辐射损伤中的治疗作用以及相关机制。方法 利用琼脂糖凝胶电泳法和原位末端标记法检测rhIL -1对 8.0Gy60Coγ射线照射小鼠小肠上皮细胞凋亡的影响,通过免疫组织化学的方法观察Bcl -和Bax的表达变化。结果 rhIL -1治疗 和预防给药可明显地抑制小鼠小肠上皮细胞凋亡的发生,并诱导Bcl 2表达而使Bax表达明显减弱。结论 rhIL-11可能通过影响 Bcl -和Bax的表达而抑制辐射引起的小肠上皮细胞凋亡,从而产生防护作用。
Objective To investigate the effects of rhIL-1 on the apoptosis of small intestine epithelial cells and the expression of related genes in irradiated mice, and to explore the therapeutic effect of IL-11 on the radiation damage of small intestinal epithelial cells in acute radiation sick mice. Methods The effects of rhIL-1 on the apoptosis of small intestine epithelial cells of mice exposed to 8.0Gy 60Coγ ray were detected by agarose gel electrophoresis and in situ end labeling. The expressions of Bcl - and Bax were observed by immunohistochemistry. Results The treatment with rhIL-1 and the prophylaxis of rhIL-1 could obviously inhibit the apoptosis of mouse intestinal epithelial cells and induce the expression of Bcl-2, which markedly decreased the expression of Bax. Conclusion rhIL-11 may inhibit the apoptosis of intestinal epithelial cells induced by radiation through affecting the expression of Bcl - and Bax, thus resulting in the protective effect.
