在整体状态下观察P物质对家兔心脏功能的影响。静脉注射3.7×10-7mol/LP物质1ml/Kg后,反映心脏功能的一些指标发生明显改变。心肌收缩成分最大缩短速率(Vmax)、左室收缩峰压(LVSP)、左室内压最大变化速率(dp/dtmax)等显著下降,左室开始收缩至发生dp/dtmax的间隔时间(t-dp/dtmax)和左室射血时间(ET)延长,左室终未舒张压(LVEDP)显著升高;心率(HR)略有减慢,而心输出量(CO)和心指数(CI)则显著升高。代表心肌耗氧的TTI及左室做功指数(LVWI)在给药后显著降低。用3.7×10-6mol/LP物质阻断剂(D—SP)1ml/kg预处理后,上述作用均可被阻断。结果表明:SP经SP受体的介导,能抑制心肌收缩性能,即对心脏有负性变力作用。
