本文应用国产全反式RA对10例APL患者的白血病细胞进行了体外诱导分化试验.结果发现,RA处理组培养7天后,89%细胞在形态上接近成熟粒细胞:NBT阳性细胞明显增高,细胞化学染色表明诱导分化细胞向过渡或成熟中性粒细胞分化.经RA治疗后,所有病例L-CFU形成明显低落,正常造血祖细胞集落大幅度上升.上述观察证实了全反式维甲酸能诱导APL患者的早幼粒细胞沿着粒系方向分化成熟而实现CR.
In this study, the leukemic cells of 10 APL patients were induced and differentiated in vitro using domestic all-trans-RA, and the results showed that 89% of the cells in morphology were close to the mature granulocytes: the NBT-positive cells were significantly increased, Chemical staining showed differentiation of differentiated cells into transitional or mature neutrophils.After treatment with RA, the formation of L-CFU in all cases was significantly reduced and the hematopoietic progenitor colonies increased significantly.The above observations confirmed that all-trans retinoic acid can be induced The promyelocytic cells of APL patients differentiate and mature along the granulocyte direction to achieve CR.
