目的探讨自发性2型糖尿病大鼠体内脂联素(ADP)水平对肝脏脂代谢的影响及其分子机制。方法自发性2型糖尿病大鼠和同系健康大鼠各20只分别作为自发性2型糖尿病组和对照组,均给予普通饲料喂养,于32周处死,处死前禁食8 h,测定大鼠空腹血糖(FPG),血浆甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(FFA)、脂联素(ADP)水平和空腹胰岛素水平,肝组织TC和TG含量,应用蛋白免疫印迹技术检测肝组织内腺苷酸活化蛋白激酶(AMPK)及磷酸化腺苷酸活化蛋白激酶(p-AMPK)蛋白水平。结果自发性2型糖尿病组大鼠FPG、血浆TC、TG、FFA和空腹胰岛素水平[分别为(6.50±2.10)、(1.82±0.36)、(0.75±0.05)mmol/L,(0.80±0.15)μmol/L,(3.35±0.45)mIU/L]均高于对照组[分别为(4.80±1.10)、(1.79±0.30)、(0.45±0.06)mmol/L,(0.45±0.09)μmol/L,(1.45±0.16)mIU/L];ADP水平[(1.49±0.08)μg/ml]明显低于对照组[(2.79±0.26)μg/ml];肝脏p-AMPK表达量(0.65±0.07)明显低于对照组(1.11±0.08),差异均有统计学意义(P<0.01)。结论 ADP可能通过AMPK影响肝脏胰岛素信号转导及糖脂代谢,参与2型糖尿病的发生发展。
Objective To investigate the effect of adiponectin (ADP) on hepatic lipid metabolism and its molecular mechanism in spontaneously type 2 diabetic rats. Methods 20 spontaneously type 2 diabetic rats and 20 healthy homologous rats were used respectively as spontaneous type 2 diabetes mellitus group and control group. All rats were fed with normal diet and sacrificed at 32 weeks. Fasting rats were fasted for 8 hours before sacrifice. FPG, TG, TC, FFA, ADP and fasting insulin, liver tissue TC and TG contents were determined by Western blotting Adenosine Activated Protein Kinase (AMPK) and phosphorylated adenylate activated protein kinase (p-AMPK) in liver tissue. Results FPG, plasma TC, TG, FFA and fasting insulin levels were significantly higher in spontaneous type 2 diabetic rats [(6.50 ± 2.10), (1.82 ± 0.36), (0.75 ± 0.05) mmol / L and (0.80 ± 0.15) (4.80 ± 1.10), (1.79 ± 0.30), (0.45 ± 0.06) mmol / L, (0.45 ± 0.09) μmol / L (1.45 ± 0.16) mIU / L]; ADP level [(1.49 ± 0.08) μg / ml] was significantly lower than that of the control group [(2.79 ± 0.26) μg / Significantly lower than that of the control group (1.11 ± 0.08), the difference was statistically significant (P <0.01). Conclusion ADP may be involved in the occurrence and development of type 2 diabetes through AMPK affecting hepatic insulin signal transduction and glycolipid metabolism.
