目的 :探讨补肾活血中药对乳腺萎缩、增生雌性大鼠乳腺组织形态结构的影响及对受体水平的影响机理。方法 :分别用去势和长期大量肌肉注射己烯雌酚的方法建立成年大鼠乳腺萎缩和乳腺增生模型 ,探讨补肾活血中药对乳腺重量指数、乳腺组织结构及乳腺雌、孕激素受体的影响。结果 :(1)补肾活血中药能使萎缩的乳腺组织结构得到明显改善 ,重量指数显著增加 (P <0 0 1) ,乳腺组织胞浆和胞核雌、孕激素受体的数量明显升高 (P <0 0 1,P <0 0 5) ,并使核 /浆雌、孕激素受体比值趋于正常。 (2 )补肾活血中药能够使增生乳腺的重量指数及乳腺结构得到恢复或改善 ,使雌、孕激素核 /浆受体比值均趋于正常。结论 :(1)补肾活血中药能够预防与改善性激素水平低下导致的成年大鼠乳腺萎缩 ,并促进大鼠乳腺发育。 (2 )补肾活血中药与雌激素制剂虽然都具有促进乳房发育的作用 ,但作用途径不同。补肾活血中药治疗乳房发育不良具有应用开发前景。
Objective : To investigate the effect of Bushen Huoxue Chinese medicine on the morphological structure of mammary gland tissue in atrophic and hyperplastic rat mammary gland and its influence mechanism on receptor level. METHODS: Adult rat mammary atrophy and mammary gland hyperplasia models were established by ovariectomized and long-term intramuscular injection of diethylstilbestrol. The effects of Bushen Huoxue herbs on mammary gland weight, mammary gland structure, and estrogen and progesterone receptors in mammary glands were investigated. Results: (1) Bushen Huoxue herbs can significantly improve the atrophic mammary gland structure, increase the weight index significantly (P < 0.01), increase the number of estrogen and progesterone receptors in cytoplasm and nucleus of mammary gland tissue ( P <0 0 1, P <0 0 5), and the nuclear/plasma estrogen and progesterone receptor ratios became normal. (2) Kidney-tonifying and blood-activating Chinese herbs can restore or improve the weight index and mammary gland structure of hyperplastic mammary glands, and the ratio of nuclear and plasma receptors of estrogen and progesterone tends to be normal. Conclusion: (1) Bushenhuoxue can prevent and ameliorate mammary atrophy in adult rats caused by low sex hormone levels and promote mammary gland development in rats. (2) Although both kidney-enhancing and blood-activating Chinese medicine and estrogen preparations have the effect of promoting breast development, the way of action is different. Bushen Huoxue Chinese medicine treatment of breast dysplasia has application development prospects.
