目的探讨皮肤鳞状细胞癌(鳞癌)组织中Notch1,NF-κB p65mRNA和蛋白的表达及其与临床病理参数的关系。方法应用RT-PCR和免疫组化SP法分别检测58例皮肤鳞癌组织及58例正常皮肤组织中Notch1及NF-κBp65mRNA和蛋白的表达。结果皮肤鳞癌组织中Notch1mRNA相对含量(0.451±0.072)低于其在正常皮肤组织中的表达(0.715±0.085),组间比较差异有统计学意义(P<0.05);NF-κBp65mRNA相对含量(0.646±0.062)高于其在正常皮肤组织中的表达(0.445±0.063),组间比较差异有统计学意义(P<0.05)。皮肤鳞癌组织中Notch1蛋白阳性表达率31.03%(18/58)低于正常皮肤组织77.59%(45/58),组间比较差异有统计学意义(P<0.05);而NF-κBp65蛋白阳性表达率81.03%(47/58)高于正常皮肤组织34.48%(20/58),组间比较差异有统计学意义(P<0.05)。结论 Notch1的低表达和NF-κBp65的过表达可能与皮肤鳞癌的发生发展密切相关。
Objective To investigate the expression of Notch1, NF-κB p65 mRNA and protein in skin squamous cell carcinoma (SCC) and its relationship with clinicopathological parameters. Methods The expressions of Notch1 and NF-κBp65 mRNA and protein in 58 cases of squamous cell carcinoma and 58 cases of normal skin tissues were detected by RT-PCR and immunohistochemistry respectively. Results The relative content of Notch1 mRNA (0.451 ± 0.072) in skin squamous cell carcinoma tissues was lower than that in normal skin tissues (0.715 ± 0.085), the difference was statistically significant (P <0.05); The relative content of NF-κBp65 mRNA 0.646 ± 0.062) was higher than that in normal skin tissues (0.445 ± 0.063). The difference between the two groups was statistically significant (P <0.05). The positive rate of Notch1 protein in skin squamous cell carcinoma was 31.03% (18/58), which was lower than that in normal skin tissue (77.59%, 45/58), the difference was statistically significant (P <0.05); while the positive expression of NF-κBp65 The expression rate of 81.03% (47/58) was higher than that of normal skin tissue (34.48%, 20/58). The difference between the two groups was statistically significant (P <0.05). Conclusion The low expression of Notch1 and the overexpression of NF-κBp65 may be closely related to the occurrence and development of squamous cell carcinoma.
