目的 将淫羊藿苷 (icariin ,ICA )和济南假单胞菌制剂 (pseudomonasjinanensisagent ,PJA)作用于人高转移肺癌细胞PG ,从肿瘤转移抑制的多个方面和环节探讨两药抗转移作用的机制 ,并观察其联合用药的作用性质。方法 采用粘附实验、运动侵袭实验、流式细胞术、细胞免疫组化等多种方法进行了检测 ,并按金正均的Q值法计算联合用药效果。结果 ICA、PJA可明显降低PG细胞对层粘连蛋白基质的粘附率 (P <0 0 5 ,P <0 0 5 ) ,并且这种粘附抑制作用有时间依赖性 ;经ICA 2 0 0mg·L-1、PJA 0 35mg·L-1及ICA 2 0 0mg·L-1+PJA 0 35mg·L-1处理 48h后 ,侵袭细胞数由对照组的 470± 5 8分别降为 2 37± 31(P <0 0 1)、316± 34(P <0 0 5 )、199± 2 3(P <0 0 1) ;运动细胞数由对照组的 5 2 0±5 6 ,分别降为 35 4± 5 5 (P <0 0 5 )、375± 19(P <0 0 5 )、2 5 6± 2 2 (P <0 0 1) ,并同时伴有细胞表面粘附分子CD4 4V6、LN R及胞浆内CK18表达的减少。其中两药联合处理对粘附、运动、侵袭能力的抑制作用均为单纯相加效果。药物处理 96h后 ,细胞表面HLA ABC抗原表达率由对照组的10 78%分别升高至 37 5 4%、38 88%、5 0 75 % (P <0 0 5 )。结论 ICA、PJA通过对肿瘤转移多个步骤的抑制而发挥抗转移作用?
Objective To treat icariin (ICA) and Pseudomonas jinanensis agent (PJA) on human high metastatic lung cancer cell line PG, and explore the mechanism of anti-metastasis effect of both drugs from several aspects and links of tumor metastasis inhibition. , And observe the role of its combination of drugs. Methods Adhesion assays, exercise invasion assays, flow cytometry, and cell immunohistochemistry were used for detection. The effect of combination therapy was calculated by Kim Jong-soon’s Q-value method. Results ICA and PJA significantly decreased the adhesion rate of PG cells to the laminin matrix (P < 0.05, P <0.05), and the inhibition of this adhesion was time-dependent; ICA 2.0 mg. After treatment with L-1, PJA 0 35 mg·L -1 and ICA 200 mg·L -1 + PJA 0 35 mg·L -1 for 48 h, the number of invading cells decreased from 470 ± 5 8 in the control group to 2 37 ± 31. The inhibitory effects of the two drugs combined treatment on adhesion, motion, and invasive ability were all simple additive effects. After 96 hours of drug treatment, the expression of HLA ABC antigen on the cell surface increased from 10 78% in the control group to 37 54%, 38 88%, and 5070%, respectively (P < 0 05). Conclusion ICA and PJA exert anti-metastatic effects by inhibiting multiple steps of tumor metastasis.
