目的建立人血清中硬脂酸(C18∶0)、油酸(C18∶1)、亚油酸(C18∶2)、十八碳三烯酸(C18∶3)、二十碳四烯酸(C20∶4)、二十碳五烯酸(EPA,C20∶5)和二十二碳六烯酸(C22∶6)7种饱和及不饱和脂肪酸的气相色谱测定方法。方法人血清中的7种脂肪酸经10%硫酸-甲醇在60℃水浴30 min,甲酯化。取出放冷至室温,加入正己烷和饱和氯化钠溶液,旋涡混匀,离心后取上层正己烷相,用气相色谱法测定,用相应的脂肪酸甲酯定性,外标法定量。结果在优化好的色谱条件下,7种脂肪酸甲酯化后可完全分离,重复性好,在5.0 mg/L~100 mg/L内相关系数(r)>0.999 4,检出限≤1.2 mg/L,10 mg/L、20 mg/L、50 mg/L 3个浓度的加标平均回收率为86.7%~93.6%,RSD为1.3%~3.2%。结论本方法前处理简单,定量准确,适用于人血清中饱和与不饱和脂肪酸的同时测定。
OBJECTIVE To establish a method for the determination of stearic acid (C18:0), oleic acid (C18:1), linoleic acid (C18:2), octadecatrienoic acid (C18: 3) C20: 4), eicosapentaenoic acid (EPA, C20: 5) and docosahexaenoic acid (C22: 6) were determined by gas chromatographic method of seven kinds of saturated and unsaturated fatty acids. Methods Seven kinds of fatty acids in human serum were methylated by 10% sulfuric acid-methanol at 60 ℃ for 30 min. Remove and let cool to room temperature, adding n-hexane and saturated sodium chloride solution, vortex mixing, after centrifugation to take the upper n-hexane phase, determined by gas chromatography, with the corresponding fatty acid methyl ester qualitative, external standard quantitative. Results Under optimized chromatographic conditions, the seven fatty acids were completely separated after methyl esterification. The correlation coefficient (r)> 0.999 4 was 5.0 mg / L ~ 100 mg / L with the detection limit of ≤ 1.2 mg The average recoveries for the three concentrations of 10 mg / L, 20 mg / L and 50 mg / L were 86.7% -93.6% and RSDs were 1.3% -3.2%. Conclusion The method is simple and accurate in pre-treatment and is suitable for the simultaneous determination of saturated and unsaturated fatty acids in human serum.
