目的:检测小鼠应用桂枝浸出物后腹腔巨噬细胞内TNFα-mRNA含量的变化。方法:雌雄小鼠随机分组,口服给药后制备巨噬细胞涂片。克隆人TNF-DNA的PGEM-1质料转化EcoliJM103,选择培养扩增。随机引物法制备地高辛标记的探针,利用原位杂交技术检测巨噬细胞内TNFα-mRNA的表达。结果:抗肿瘤中草药桂枝等浸出物可增强小鼠腹腔巨噬细胞内TNFα-mRAN-mRAN表达。结论:分子生物学的方法与手段可用于揭示抗肿瘤中草药的作用机理。
OBJECTIVE: To detect the changes of TNFα-mRNA content in peritoneal macrophages after the application of cassia twig extract. METHODS: Male and female mice were randomized and macrophage smears were prepared after oral administration. The PGEM-1 material from which human TNF-DNA was cloned was transformed into EcoliJM103 and selected for expansion. Digoxigenin-labeled probes were prepared by random primer method and the expression of TNFα-mRNA in macrophages was detected by in situ hybridization. RESULTS: Anti-tumor herbs such as cassia twigs could enhance the expression of TNFα-mRAN-mRAN in mouse peritoneal macrophages. Conclusion: Molecular biology methods and methods can be used to reveal the mechanism of action of anti-tumor Chinese herbs.
