目的探讨大鼠纳米硫化铅(PbS NPs)染毒后,全血、心、肝、肺、肾和骨组织内锌、铁、铅含量的变化。方法 SPF级Wistar雄性大鼠19只,按体质量随机分为高剂量组(30mg/kg,7只),低剂量组(15mg/kg,6只)和对照组(0mg/kg,6只)。其中高、低剂量组分别经呼吸道灌注粒径为20nm PbS混悬液1ml,对照组灌注等体积的生理盐水,每周1次,连续3个月;最后1次染毒后24h处死大鼠,心脏取血后,取其心、肝、肺、肾和骨组织。采用电感耦合等离子体质谱仪(ICP-MS)测定各组织中铅、锌、铁元素的含量。结果 PbS NPs染毒大鼠全血高剂量组、心、肝、肺、肾和骨组织的高、低剂量组铅含量均明显高于对照组,差异有统计学意义(P<0.05);而全血低剂量组铅含量与对照组比较,差异无统计学意义(P>0.05);PbS NPs组大鼠体全血和机体各组织中的锌、铁元素含量分别与对照组比较,差异有统计学意义(P<0.05)。结论 PbS NPs暴露可能导致大鼠全血、心、肝、肺、肾和骨组织发生铅蓄积;全血和机体各组织中锌、铁元素含量降低。
Objective To investigate the changes of zinc, iron and lead contents in whole blood, heart, liver, lung, kidney and bone after exposure to PbS NPs. Methods Nineteen SPF Wistar male rats were randomly divided into high dose group (30mg / kg), low dose group (15mg / kg, 6 mice) and control group (0mg / kg, . The high and low dose groups were respectively infused with 1ml PbS suspension of 20nm in diameter through the respiratory tract, and the control group was infused with equal volume of normal saline once a week for 3 months. Rats were sacrificed 24 hours after the last injection, After taking the heart blood, take its heart, liver, lung, kidney and bone tissue. The contents of lead, zinc and iron in each tissue were determined by inductively coupled plasma mass spectrometry (ICP-MS). Results The levels of lead in high and low dose groups of heart, liver, lung, kidney and bone in PbS NPs-treated high-dose whole blood group were significantly higher than those in control group (P <0.05) There was no significant difference in lead content between low-dose and low-dose whole blood groups (P> 0.05). The contents of zinc and iron in whole blood and tissues of rats in PbS NPs group were significantly lower than those in control group Statistical significance (P <0.05). Conclusion Exposure of PbS NPs may lead to lead accumulation in whole blood, heart, liver, lung, kidney and bone in rats. The content of zinc and iron in whole blood and tissues of the body decreased.
