目的:观察碱性成纤维细胞生长因子(bFGF)对血管平滑肌细胞(VSMC)connexin43蛋白(Cx43)表达的影响。方法:将组织贴块法培养的兔胸主动脉VSMC随机分为空白组和实验组(bFGF浓度为3μg/L),应用免疫荧光组织化学技术检测connexin43表达。用Leica激光共聚焦显微镜观察并拍照,图片用Silicon Graphics Octane进行处理。结果:免疫荧光显示,正常VSMC中Cx43蛋白主要分布于细胞中间部和周边部。bFGF作用24 h后,VSMCs数目多于对照组,Cx43蛋白荧光强度明显高于对照组,且在细胞连接处增多。结论:bFGF不仅刺激成纤维细胞的分裂增殖,对血管平滑肌细胞Cx43蛋白表达有明显促进作用,揭示间隙连接表达的增多,可能对损伤后血管重塑起一定的作用。
Objective: To investigate the effect of basic fibroblast growth factor (bFGF) on the expression of connexin43 protein (Cx43) in vascular smooth muscle cells (VSMCs). Methods: VSMCs of thoracic aorta were randomly divided into blank group and experimental group (bFGF concentration of 3μg / L). The expression of connexin43 was detected by immunofluorescence histochemistry. Observed and photographed with a Leica laser confocal microscope and processed with Silicon Graphics Octane. Results: The results of immunofluorescence showed that Cx43 protein in normal VSMCs mainly distributed in the middle and peripheral part of cells. After bFGF treatment for 24 h, the number of VSMCs was more than that of the control group. The fluorescence intensity of Cx43 protein was significantly higher than that of the control group, and increased at the cell junction. CONCLUSION: bFGF not only stimulates the proliferation and proliferation of fibroblasts but also promotes the expression of Cx43 protein in vascular smooth muscle cells. It reveals that the increase of gap junctional expression may play a role in vascular remodeling after injury.
