以甘蓝型油菜92P183试管苗为材料,以B5基本培养基添加生长素类物质NAA、IAA、IBA及其组合作为处理,筛选适合甘蓝型油菜试管苗生根的最佳培养基。结果表明,10种处理的培养基,其生根效果差异显著,以IBA0.5mg/L的处理效果最佳。接种后6~7d第1株试管苗生根,8d50%试管苗生根。培养25d统计,生根率100%,平均根数11.22条,平均根长度6.99cm。第1株试管苗生根后的2~4d内,生根苗数达到最高峰,这时1d的生根苗数几乎占总接种苗数的1/3。
The optimum culture medium for the rooting of in vitro Brassica napus seedlings was screened by using NAA, IAA, IBA and their combinations of auxin NAA, IAA and IBA in 92P183 test tube seedlings of Brassica napus. The results showed that the rooting effect of the 10 kinds of medium was significantly different, IBA0.5mg / L the best treatment. After inoculation 6 ~ 7d first plantlets rooting, 8d50% of plantlets rooting. Training 25d statistics, rooting rate of 100%, the average number of 11.22, the average root length of 6.99cm. The number of rooting seedlings peaked at 2 ~ 4 days after rooting of the first plantlets in vitro.
