目的:观察蝎毒抗癌多肽(antineoplasticpolypeptidefromButhusMatensivenom,APBMV)对人肿瘤细胞生长的抑制作用。方法:采用噻唑蓝(MTT)法、台盼蓝拒染法和集落形成观察为指标,丝裂霉素C(MMC)为阳性对照药品,以APBMV4mg/L,8mg/L,12mg/L和16mg/L处理4种人肿瘤细胞(人早幼粒白血病细胞株HL60、人肝细胞癌细胞株SMMC7721、人低分化鼻咽上皮癌细胞株CNE2Z和人胃癌细胞株MGC803),选用不同的作用时间点,分别观察APBMV对上述细胞的影响。结果:APBMV对HL60、SMMC7721、CNE2Z和MGC803细胞均有显著的细胞毒性作用,HL60和SMMC7721比后两者显示出更好的量效关系(r=0.9828,r=0.9863,P=0.02)。APBMV处理HL60和SMMC7721细胞24h,48h和96h后,两种细胞的生长均受到抑制(P<0.05,P<0.01),并有显著的剂量效应关系;处理SMMC7721细胞24h,48h和96h时APBMV的IC50分别是15.87mg/L、13.05mg/?
OBJECTIVE: To observe the inhibitory effect of antineoplasticpolypeptide from Brugus Matanii (APBMV) on the growth of human tumor cells. Methods: MTT assay, trypan blue exclusion assay and colony formation assay were used as indexes. Mitomycin C (MMC) was used as positive control drug.
