目的 :研究我国广西抗蛇毒中草药红背丝绸 [Cissusassamica (Laws.)Craib]对内皮素 1(endothelin 1,ET 1)的拮抗作用。方法 :对红背丝绸进行提取分离 ,用不同提取物拮抗ET 1引起的小鼠急性死亡、大鼠离体血管条收缩和整体大鼠升压作用。结果 :红背丝绸乙醇提取物、乙醇提取物之乙酸乙酯萃取物以及从乙酸乙酯萃取物中分离得到的白藜芦醇 (resveratrol)在整体和离体水平对ET 1均具拮抗作用。该 3种提取物对ET 1致小鼠急性死亡有明显的保护作用 ,亦可明显地拮抗由ET 1引起的离体大鼠主动脉条的收缩并呈剂量依赖性 ,并且能够拮抗ET 1导致的大鼠血压的上升幅度。结论 :红背丝绸及其有效成分白藜芦醇对ET 1具有生物效应拮抗作用 ,提示从某些抗蛇毒中草药寻找ET拮抗剂是可行的
Objective : To study the antagonistic effect of Cissus assamica (Laws) Craib on the endothelin-1 (ET 1) in Chinese herbal medicine, anti-venom poisoning in Guangxi, China. METHODS : Red-back silk was extracted and separated. Different extracts were used to antagonize the acute death of mice induced by ET-1, the contraction of isolated rat vascular strips and the whole-body pressure-raising effect of rats. RESULTS: Red-backed silk ethanol extracts, ethyl acetate extracts of ethanol extracts, and resveratrol isolated from ethyl acetate extracts were antagonistic to both ET 1 and total levels. The three extracts have obvious protective effects on acute death of mice induced by ET 1, and can also significantly antagonize the contraction of rat aortic strips induced by ET 1 in a dose-dependent manner, and can antagonize ET 1 The increase in blood pressure in rats. Conclusion: Red-backed silk and its active ingredient resveratrol have biological effect antagonism on ET 1, suggesting that it is feasible to search for ET antagonists from some anti-venom Chinese herbal medicines.
