目的:正交优选岩黄连的最佳提取工艺。方法:以岩黄连中总生物碱、脱氢卡维丁的含量为指标,比较乙醇和酸水两种不同提取溶媒以及回流、渗漉两种不同提取方法并采用L_9(3~4)正交设计,对影响岩黄连提取工艺中的溶剂浓度、溶剂用量、提取时间、提取次数四个因素进行优化筛选。结果:乙醇提取优于酸水提取,回流法提取效果优于渗漉法。醇提最佳工艺条件为A_2B_2C_3D_2,即75%乙醇提取,每次加醇量10倍,提取3次,每次提取时间2 h。结论:优选得到的工艺简便、稳定可行。
Objective: Orthogonal optimization of optimal extraction process of Coptis chinensis. Methods: The contents of total alkaloids and dehydrocavageine in the genus Ichthyrium were used as indexes to compare two different extraction methods of ethanol and acid water and different extraction methods of refluxing and percolation. L_9 (3 ~ 4) orthogonal Design, optimize the screening of the four factors that affect the solvent concentration, solvent dosage, extraction time, extraction times in the extraction process of Yanhuang Huanglian. Results: Ethanol extraction was better than acid water extraction, and refluxing method was better than percolation method. The optimum conditions of alcohol extraction were A_2B_2C_3D_2, which was extracted with 75% ethanol, the amount of alcohol was increased by 10 times and extracted three times, each extraction time was 2 h. Conclusion: The optimized process is simple, stable and feasible.
