长链非编码RNA为长度大于200nt的非编码RNA,具有组织表达特异性,且表达丰度较mRNA低。在前期肉牛肌肉和脂肪组织的lncRNA组学测序分析中筛查并获得一个长度为755nt的差异表达长链非编码RNA,将其命名为lncRNA_(TCONS-585)。经基因组序列比对,该novel lncRNA_(TCONS-585)序列位于牛第5号染色体MCHR1和PMP34两个基因之间,由两个外显子构成。对lncRNA_(TCONS-585)结构分析后通过RT-PCR验证了lncRNA_(TCONS-585)在靶向组织中的表达水平以及在不同组织中的表达谱。结果表明,lncRNA_(TCONS-585)在牛的不同组织中表达量具有较大差异,其在肌肉组织中表达水平最高,同时在白色脂肪和棕色脂肪组织当中也有表达,且表达量显著高于肝、肺和肾。同时,我们发现干扰lncRNA_(TCONS-585)可降低其邻近基因MCHR1的表达量。因此lncRNA_(TCONS-585)可能作为调控因子通过介导靶基因参与肉牛生长发育和能量代谢过程。
Long-chain non-coding RNA is more than 200nt length of non-coding RNA, with tissue-specific expression, and the expression of mRNA lower than mRNA. In a preliminary lncRNA sequencing analysis of muscle and adipose tissue of beef cattle, a 755-nt differentially expressed long-chain non-coding RNA was screened and named lncRNA_ (TCONS-585). The novel lncRNA_ (TCONS-585) sequence is located between the two genes of MCHR1 and PMP34 on the bovine chromosome 5 by genomic sequence alignment and consists of two exons. The lncRNA_ (TCONS-585) expression level in target tissues and the expression profiles in different tissues were verified by RT-PCR analysis of lncRNA_ (TCONS-585) structure. The results showed that the expression of lncRNA_ (TCONS-585) was significantly different in different tissues of cattle. The expression level of lncRNA_ (TCONS-585) was the highest in muscle tissue and was also expressed in white adipose tissue and brown adipose tissue, , Lungs and kidneys. At the same time, we found that interference with lncRNA_ (TCONS-585) can reduce the expression of its adjacent gene MCHR1. Therefore, lncRNA_ (TCONS-585) may play a role as a regulatory factor in the process of beef cattle growth and development and energy metabolism by mediating target genes.
