目的:探讨槲皮素对乳腺癌MCF-7细胞株热休克蛋白70(HSP70)、HSP27和HSP90的表达的影响。方法:对体外培养的乳腺癌MCF-7细胞,恒温42℃热处理2h,分别于4、8、12和24h后荧光定量PCR检测HSP70、HSP27和HSP90的基因表达变化并用蛋白质印迹检测蛋白表达量的变化。观察热处理前使用150μmol/L槲皮素对乳腺癌MCF-7细胞HSP70、HSP27和HSP90的基因表达变化、蛋白表达量的影响。结果:42℃热处理后乳腺癌MCF-7细胞HSP70、HSP27和HSP90表达在基因及蛋白水平上均有明显升高,4h达到高峰,8h后开始减少,24h接近正常值。槲皮素能抑制HSP70、HSP27表达,但导致HSP90表达水平先升高,4~8h表达仍处于高峰值,8h后才开始下降。结论:HSP70、HSP27和HSP90在MCF-7细胞内的基因和蛋白表达均在热处理后迅速升高,在热处理前使用槲皮素可以抑制HSP70、HSP27的表达,HSP90水平升高及下降延迟。
Objective: To investigate the effect of quercetin on the expression of heat shock protein 70 (HSP70), HSP27 and HSP90 in breast cancer cell line MCF-7. Methods: The MCF-7 breast cancer cells cultured in vitro were heat-treated at 42 ℃ for 2 h. The mRNA expressions of HSP70, HSP27 and HSP90 were detected by real-time PCR after 4, 8, 12 and 24 h respectively. Western blotting was used to detect the protein expression Variety. The effect of 150μmol / L quercetin on the gene expression and protein expression of HSP70, HSP27 and HSP90 in breast cancer MCF-7 cells was observed before heat treatment. RESULTS: The expression of HSP70, HSP27 and HSP90 in breast cancer MCF-7 cells after heat-treatment at 42 ℃ were significantly increased at both gene and protein levels, reaching the peak at 4h, decreasing at 8h and approaching normal at 24h. Quercetin could inhibit the expression of HSP70 and HSP27, but the expression of HSP90 first increased, and reached a peak at 4 ~ 8h, and then began to decline after 8h. CONCLUSIONS: The expression of HSP70, HSP27 and HSP90 in MCF-7 cells both increased rapidly after heat treatment. Quercetin used before heat treatment can inhibit the expression of HSP70 and HSP27, increase the level of HSP90 and delay the decline.
