采用含有4个碳原子臂长的PCB37半抗原衍生物合成PCB37全抗原,以人工抗原免疫BALB/C小鼠,通过鼠-鼠杂交瘤技术制备抗PCB37单克隆抗体。建立间接竞争ELISA(ic-ELISA)分析方法,对抗PCB37单克隆抗体的特性进行评估,最终获得了一株能够稳定分泌抗PCB37单克隆抗体的细胞株。使用该抗体建立了ic-ELISA,在PCB37浓度为0.10~50μg/L时,抑制率与PCB37的浓度呈线性关系,半数抑制浓度(IC50)为1.23μg/L,最低检测限(IC15)为0.027μg/L,与其他物质的交叉反应率<5.5%。结果表明,成功研制了抗PCB37单克隆抗体,为进一步建立检测PCB37的免疫分析检测方法奠定了基础。
The PCB37antigen was synthesized by using PCB37 hapten with 4 arms of arm length. BALB / C mice were immunized with artificial antigen and anti - PCB37 monoclonal antibody was prepared by murine - mouse hybridoma technique. The establishment of an indirect competitive ELISA (ic-ELISA) analysis method to evaluate the characteristics of the monoclonal antibody against PCB37 finally obtained a cell strain capable of stably secreting the anti-PCB37 monoclonal antibody. The concentration of PCB37 was linear with the concentration of PCB37 at the concentration of 0.10 ~ 50μg / L. The IC50 was 1.23μg / L and the IC 50 was 0.027 μg / L, cross-reactivity with other substances <5.5%. The results showed that the anti-PCB37 monoclonal antibody was successfully developed, which laid the foundation for the further establishment of an immunoassay detection method for detecting PCB37.
