目的研究分化抑制因子-1(ID-1)、血管内皮生长因子(VEGF)、凝血酶敏感蛋白-1(Tsp-1)及微血管密度(MVD)在皮肤基底细胞癌(BCC)及鳞状细胞癌(SCC)中表达的相关性。方法应用免疫组化法检查皮肤BCC及SCC各25例的手术切除标本中ID-1,VEGF,Tsp-1和微血管标记抗体CD34的表达情况,计数肿瘤MVD;应用RT-PCR方法检测ID-1,Tsp-1和VEGF的mRNA表达情况;应用Western blot方法检查ID-1,Tsp-1和VEGF蛋白水平。结果与正常皮肤相比,皮肤SCC和BCC均高表达MVD,ID-1和VEGF,低表达Tsp-1,差异均有统计学意义(P均<0.05)。结论 ID-1和VEGF与皮肤BCC及SCC组织中的血管生成正相关,二者在促进皮肤SCC和BCC血管生成中具有协同作用,并可能在皮肤SCC和BCC的发生及发展过程中起着重要作用。
Objective To investigate the expression of ID-1, VEGF, Tsp-1 and MVD in cutaneous basal cell carcinoma (BCC) and squamous cell Cancer (SCC) expression in the correlation. Methods Immunohistochemistry was used to examine the expression of ID-1, VEGF, Tsp-1 and microvessel tag antibody (CD34) in the resected specimens of 25 cases of skin BCC and SCC. The MVD of tumor was counted and the expression of ID-1 , Tsp-1 and VEGF mRNA expression were detected by Western blot, ID-1, Tsp-1 and VEGF protein levels. Results Compared with normal skin, the expression of MVD, ID-1, VEGF and low expression of Tsp-1 in skin SCC and BCC were significantly different (all P <0.05). Conclusion ID-1 and VEGF are positively correlated with angiogenesis in skin BCC and SCC tissues. Both of them have synergistic effects in promoting SCC and BCC angiogenesis in skin and may play an important role in the occurrence and development of SCC and BCC in skin effect.
