目的 研究银杏苦内酯B(BN 5 2 0 2 1)对豚鼠心室肌细胞动作电位 (AP)和L 型钙通道的影响。方法 全细胞膜片箝技术。AP记录采用电流箝方式 ,电流记录采用电压箝方式。结果 BN 5 2 0 2 1明显缩短动作电位时程 (APD) ,在10 -6mol·L-1浓度 ,APD90 缩短 9% (P <0 0 5 )。在 10 -5mol·L-1浓度 ,APD90 缩短 12 % (P <0 0 1)。 10 -6mol·L-1以上浓度BN 5 2 0 2 1还明显缩短APD50 ,最大缩短达 14% (P <0 0 5 )。较高浓度 (10 -5mol·L-1)的BN 5 2 0 2 1可使静息电位增加 (P <0 0 5 )。BN 5 2 0 2 1浓度依赖性减少L 型钙电流(L ICa)。 10 -6mol·L-1浓度下 ,峰值L ICa降低 2 4 7% (P<0 0 1) ,10 -5mol·L-1浓度下 ,峰值L ICa降低 36 9% (P <0 0 1)。随着药物浓度的增加 ,I U关系曲线逐渐上移 ,但其峰值电压保持不变。结论 BN 5 2 0 2 1明显缩短APD ,抑制L ICa,且具有明显的浓度依赖关系。
Objective To study the effects of Ginkgolide B (BN 5221) on action potential (AP) and L-type calcium channels in guinea pig ventricular myocytes. Methods Whole cell patch clamp technique. The AP record uses the current clamp mode and the current record uses the voltage clamp mode. Results BN 52 1 2 significantly shortened the action potential duration (APD), at 10 -6 mol·L -1 , and APD90 decreased by 9% (P <0 05). At 10 -5 mol·L-1, APD90 was shortened by 12% (P < 0 01). The concentration of BN 5 2 0 2 1 above 10 -6 mol·L-1 also significantly shortened the APD50, which was shortened by up to 14% (P <0 05). A higher concentration (10 -5 mol·L-1) of BN 5 2 0 2 1 increased the resting potential (P 0 05). BN 5 2 0 2 1 concentration-dependently reduces L-type calcium current (L ICa). At a concentration of 10 -6 mol·L-1, the peak L ICa decreased by 24.7% (P<0 01), and at a concentration of 10 -5 mol·L-1, the peak L ICa decreased by 36 9% (P < 0 0 1). . As the drug concentration increases, the I U curve gradually shifts upward, but its peak voltage remains unchanged. Conclusion BN 5221 significantly shortens APD, inhibits L ICa, and has a significant concentration-dependent relationship.
