目的:研究拉米夫定(3TC)联合原儿茶酸(PA)对鸭体内鸭乙肝病毒(DHBV)的抑制作用。方法:感染DHBV的北京雏鸭模型随机分为3TC组,PA组,3TC/PA合用组及模型对照组,每天灌胃给药1次,连续给药10天。分别于给药前、给药第5天、给药第10天及停药后第3天取血,检测血清中DHBV DNA、白蛋白(ALB)和总胆红素(TBil)含量,以及谷丙转氨酶(ALT)、谷草转氨酶(AST)和碱性磷酸酶(ALP)活性,并对各时间点肝组织进行病理形态学分析。结果:PA、3TC单用及合用对雏鸭血清DHBV DNA有抑制作用,且合用时的抑制作用最为显著,停药3天后仍与给药前有显著性差异。同时,3TC与PA合用组血清中ALT、AST和ALP活性,以及ALB和TBil含量均较模型对照组有一定程度的降低。另外,3TC组肝脏病理形态与模型对照组相比无明显改善,但PA及其与3TC合用组肝组织病理形态均有一定程度改善。结论:3TC与PA合用可有效抑制雏鸭的DHBV感染,且对DHBV所致雏鸭肝损伤有一定保护作用。
Objective: To study the inhibitory effect of lamivudine (3TC) combined with protocatechuic acid (PA) on duck hepatitis B virus (DHBV) in ducklings. Methods: Beijing ducklings infected with DHBV were randomly divided into 3TC group, PA group, 3TC / PA combined group and model control group. Blood samples were collected before treatment, on the fifth day, on the tenth day after drug administration and on the third day after the drug administration. The contents of DHBV DNA, albumin (ALB) and total bilirubin (TBil) (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activity were measured. Liver histopathology was analyzed at each time point. Results: PA and 3TC alone and in combination inhibited the serum DHBV DNA of ducklings, and the combination of PA and 3TC had the most significant inhibitory effect. After stopping for 3 days, there was a significant difference between before and after administration. At the same time, the activities of ALT, AST and ALP, ALB and TBil in 3TC and PA combined groups were lower than those in model control group. In addition, 3TC group liver pathological morphology and control group compared to no significant improvement, but the combination of PA and 3TC liver histopathology have improved to some extent. Conclusion: The combination of 3TC and PA can effectively inhibit DHBV infection in ducklings and protect DHFV-induced duck liver injury.
