百日咳全细胞菌苗经液体培养、酸沉淀和56℃30分钟脱毒制成.无细胞制剂是酸沉淀后经两次离心(15000g×15分钟)及磷酸盐浸出提取.以小鼠体重变化判定毒性试验结果,用百日咳杆菌作脑内攻击检测两种制剂对小鼠的保护力.将两种制剂以不同剂量接种小鼠后5天测定组胺致敏活性,计算24小时内小鼠死亡数,按Reed等方法计算结果.结果表明,6μg无细胞制剂即可产生93.75~100%的保护作用;而600μg的全细胞菌苗只能使75.00~81.25%的小鼠存活.在本实验所用的剂量范围内,两种制剂均未使小鼠的体重发生变化.两种制剂产生的组胺致敏活性呈剂量反应关系,但全细胞菌苗
