目的探讨中药有效成分穿心莲内酯(Andrographolide,AG)对白假丝酵母菌黏附作用的影响。方法采用XTT减低法分别评价AG对白假丝酵母菌早期黏附的影响;倒置显微镜观察该药对生物膜形成的早期形态学影响;采用实时荧光定量PCR法检测其黏附相关基因ALS3、EAP1、ALS1的表达量。结果 AG对白假丝酵母菌2 h的早期黏附的干预作用较明显;qRT-PCR结果表明AG可显著抑制ALS3、EAP1的表达,但对ALS1的表达具有促进作用。结论 AG可能通过抑制白假丝酵母菌某些黏附基因的表达而抑制其早期黏附。
Objective To investigate the effect of Andrographolide (AG), an effective component of traditional Chinese medicine, on the adhesion of Candida albicans. Methods The effect of AG on early adhesion of Candida albicans was evaluated by XTT reduction method. The morphological changes of early stage of biofilm formation were observed by inverted microscope. The adhesion-related genes ALS3, EAP1 and ALS1 were detected by real-time fluorescence quantitative PCR The amount of expression. Results The effect of AG on the early adhesion of Candida albicans at 2 h was obvious. The results of qRT-PCR showed that AG could significantly inhibit the expression of ALS3 and EAP1, but could promote the expression of ALS1. Conclusion AG may inhibit its early adherence by inhibiting the expression of some adhesion genes of Candida albicans.
