目的探讨钙通道阻滞剂维拉帕米对脑膜瘤细胞增殖的影响。方法将不同浓度的维拉帕米作用于培养的脑膜瘤细胞,MTT法观察其对细胞增殖的抑制作用。建立裸鼠皮下脑膜瘤模型,观察服用维拉帕米后肿瘤的生长情况,免疫组化检测皮下肿瘤增殖细胞核抗原(PCNA)的表达。结果维拉帕米浓度呈依赖性地抑制脑膜瘤细胞增殖,浓度为1μmol/L时即有抑制效应,100μmol/L时抑制作用最明显。服药组肿瘤的体积明显小于对照组(P<0.01),其PCNA的表达也低于对照组(P<0.05)。结论维拉帕米在体外和体内均可抑制脑膜瘤细胞的增殖和生长。
Objective To investigate the effect of verapamil, a calcium channel blocker, on the proliferation of meningioma cells. Methods Different concentrations of verapamil were applied to cultured meningioma cells. MTT assay was used to observe the inhibitory effect of verapamil on cell proliferation. The model of subcutaneous meningioma in nude mice was established. The growth of tumors was observed after verapamil administration. The expression of proliferating cell nuclear antigen (PCNA) in subcutaneous tumor was detected by immunohistochemistry. Results Verapamil inhibited the proliferation of meningioma cells in a concentration-dependent manner. When the concentration was 1 μmol / L, the inhibitory effect was observed. The inhibition was the most obvious at 100 μmol / L. The tumor volume of the medication group was significantly smaller than that of the control group (P <0.01), and the expression of PCNA was also lower than that of the control group (P <0.05). Conclusion Verapamil can inhibit the proliferation and growth of meningioma cells both in vitro and in vivo.
