目的:探讨大鼠海马立体定向注射β-淀粉样蛋白1-40(Aβ1-40)后血清TNF-α和IL-1β含量的变化及加减地黄饮子对其影响。方法:100只SD大鼠随机分为空白对照组、假手术对照组、模型对照组、盐酸多奈哌齐对照组、加减地黄饮子组共5组,采用海马立体定向注射Aβ1-40诱导Alzheimer’s病动物模型后,药物干预4周,第5周应用双抗夹心酶联免疫吸附测定法检测TNF-α和IL-1β含量。结果:模型组TNF-α和IL-1β含量显著高于假手术组(p<0.05)。与模型组比较,盐酸多奈哌齐组和加减地黄饮子组TNF-α和IL-1β含量显著降低(p<0.05)。结论:大鼠海马注射Aβ1-40后大鼠血清TNF-α和IL-1β含量明显增高,加减地黄饮子通过降低血清TNF-α和IL-1β的含量,从而发挥其抗老年性痴呆的作用。
OBJECTIVE: To investigate the changes of serum TNF-α and IL-1β levels in hippocampus after stereotactic injection of β-amyloid 1-40 (Aβ1-40) and to evaluate the effect of diterpenes on the content of TNF-α and IL-1β. Methods: One hundred Sprague-Dawley rats were randomly divided into blank control group, sham-operated control group, model control group, control group of donepezil hydrochloride, and addition and subtraction group of Dihuangyinzi group. Five groups of Alzheimer’s disease were induced by stereotactic injection of Aβ1-40 in the hippocampus. After the model, drug intervention for 4 weeks, 5 weeks using double-antibody sandwich enzyme-linked immunosorbent assay to detect TNF-α and IL-1β content. Results: The levels of TNF-α and IL-1β in the model group were significantly higher than those in the sham group (p<0.05). Compared with the model group, the levels of TNF-α and IL-1β in the donepezil hydrochloride group and the decoction and rehmannia decoction group were significantly decreased (p<0.05). Conclusion: The levels of serum TNF-α and IL-1β in rat hippocampus after injection of Aβ1-40 are significantly increased. The addition and subtraction of Dihuangyinzi can reduce the content of serum TNF-α and IL-1β, thus exerting its anti-senile dementia. effect.
